ABSTRACT

Objectives: The Purpose Of This Study Was To Evaluate The Effect Of Mammalian Target Of Rapamycin (mTOR) Inhibitor (everolimus) On Squamous Cell Carcinoma Cell Line.

Materials and Methods: Prepared Human Squamous Cell Carcinoma Cell Line (Hep-2) Was Treated With Different Concentrations Of Everolimus. The Effect Of Everolimus On Cell Line Was Investigated Using MTT Cytotoxicity Assay And Nuclear Morphometric Analysis

Results: The MTT Assay Showed That The Viability Of Everolimus Treated Hep-2 Cells Decreased As The Concentration Of Everolimus Increased From 8 To 500 NM With IC50 Being 132 NM. Morphometric Analysis Showed A Decrease In Nuclear Area Factor (NAF) Which Is An Indicator Of Apoptosis As Concentration Increased. Conclusion: The MTOR Inhibitor (everolimus) Has A Toxic Effect On Squamous Cell Carcinoma Cell Line.
