IMMUNOHISTOCHEMICAL EXPRESSION OF CYCLOOXGYNASE-2 AND TRANSFORMING GROWTH FACTOR-?1 IN ADENOID CYSTIC CARCINOMA OF SALIVARY GLAND (IN VITRO STUDY)

ABSTRACT
Background: ACC Has Several Distinctive Characteristics Including Multiple Local Recurrences, High Incidences Of Metastasis And Poor Prognosis. Therefore Studying Effective Markers To Predict The Risk Of Malignancy And Clarifiction Of The Underlying Molecular Mechanisms Are Essential To Understand The Clinical Behavior And Facilitate The Management Of ACC. Objective: To Investigate The Immunohistochemical Expression Of TGF-?1 And COX-2 In Different Variants Of ACC (cribriform, Tubular/trabecular And Solid) And Correlate The Expression Of The Two Markers With Each Other. Methods: A Total Of 30 Specimens Were Collected. The Standard Avidin-biotinperoxidase Technique Has Been Used In The Present Study For Detection Of TGF- ?1 And COX-2, The Immunostaining Of These Markers Were Evaluated By An Image Analyzer Computer System.

Results: COX-2 And TGF-?1expression Was Significantlyincreased In Solid Variant Of ACC Than Other Patterns And Thus Concluded That COX-2 And TGF-?1expression Are Directly Related To ACC Histopathological Grade. Also, A Significantdirect Correlation Between The Immunostaining Of TGF-?1 And COX-2 Was Found. In Most Of Cases The Increase In COX-2 Expression Is Associated With Increase In TGF-?1expression, Which May Reveal That TGF-?1 Induce The Expression Of COX-2.Conclusion: COX-2 And TGF-?1 Expression Was Highly Expressed In Solid Variant Of ACC Than In Other Variants. There Was A Significantdirect Correlation Between TGF-?1 And COX- 2; P Value = 0.001 (P-values Less Than 0.05 Were Considered As Statistically Significant)an Increase In COX-2 Expression Is Associated With Increase In TGF-?1expression In All Studied Cases.
